You have to route a MIDI track into the effect instance (via the MIDI tracks destination menu) and then you will hear sound when notes occur on the MIDI track and sound is running through the effect instance.

Okay, there is indeed a problem in Live 5 with the output always being silent and I've found the reason.  Rez Synth is most likely affected too.  I will fix it and get new releases out as soon as I can...
